Fluoride exposure during pregnancy and lactation triggers oxidative stress and molecular changes in hippocampus of offspring rats

Long-term exposure to high concentrations of fluoride (F) can damage mineralized and soft tissues such as bones, liver, kidney, intestine, nervous system adult rats. The permeability the blood–brain barrier placenta F during pregnancy lactation may be critical neurological development. Therefore, this study aimed investigate effects on molecular processes oxidative biochemistry offspring rats’ hippocampus. Pregnant Wistar rats were randomly assigned into 3 groups in accordance with drinking water received: G1 – deionized (control); G2 10 mg/L G3 50 F. fluoridated began first day lasted until 21st breastfeeding (when weaned). Blood plasma samples collected determine levels. Hippocampi for analyses through antioxidant capacity against peroxyl (ACAP), lipid peroxidation (LPO), nitrite (NO2?) Also, brain-derived neurotrophic factor (BDNF) gene expression (RT-qPCR) proteomic profile performed. results showed that both increased bioavailability, triggered redox imbalance featured by a decrease ACAP, increase LPO NO2? levels, BDNF overexpression changes hippocampus proteome. These findings raise novel questions regarding potential repercussions structure functioning different cognitive domains.
